Do soutěže mužské čtyřhry na tenisovém turnaji Ecuador Open Quito 2016 nastoupilo šestnáct dvojic. Obhájcem titulu byl německý pár Gero Kretschmer a Alexander Satschko, jehož členové skončili ve čtvrtfinále na raketách brazilské dvojice Thomaz Bellucci a Marcelo Demoliner.
Vítězem čtyřhry se stal nenasazený španělsko-argentinský pár Pablo Carreño Busta a Guillermo Durán, který ve finále zdolal brazilskou dvojici Thomaz Bellucci a Marcelo Demoliner výsledkem 7–5 a 6–4.
Oba šampioni si do žebříčku ATP připsali 250 bodů a v premiérovém finále na okruhu ATP Tour získali první titul. V následném vydání deblového žebříčku ATP z 8. února 2016 figurovali na svých kariérních maximech, když Carreñovi patřila 82. příčka a Demolinerovi 55. místo.
