Do soutěže mužské čtyřhry na tenisovém turnaji U.S. Men's Clay Court Championships 2016 nastoupilo šestnáct dvojic. Obhájcem titulu byl pár Ričardas Berankis a Teimuraz Gabašvili, jehož členové v Houstonu nestartovali. Gabašvili se rozhodl nastoupit na paralelně probíhajícím Grand Prix Hassan II v Maroku.
Vítězem čtyřhry se stal nejvýše nasazený americký pár Bob a Mike Bryanovi, který ve finále zdolal dominikánsko-mexickou dvojici Víctor Estrella Burgos a Santiago González výsledkem 4–6, 6–3 a [10–8].
Oba šampioni si do žebříčku ATP připsali 250 bodů. Získali také rekordní 110. společný titul, premiérový v probíhající sezóně i šestou trofej z Houstonu. Naposledy předtím triumfovali na srpnovém Rogers Cupu 2015 v Montréalu. Mezitím nevyhráli ani jeden z dvanácti turnajů, což byla jejich nejdelší šňůra bez trofeje od sezóny 2001–2002.[1]

Concepción Bona was helped by her first cousin María de Jesús Pina.
