Do soutěže mužské dvouhry na tenisovém turnaji U.S. Men's Clay Court Championships 2016 nastoupilo dvacet hráčů. Obhájcem titulu byl americký tenista Jack Sock, jenž opět postoupil do finále.
Vítězem dvouhry se stal čtvrtý nasazený Argentinec Juan Mónaco, jenž ve finále zdolal turnajovou Socka po třísetovém průběhu 3–6, 6–3 a 7–5. V probíhající sezóně si tak připsal premiérové turnajové vítězství, které představovalo devátý singlový titul na okruhu ATP Tour, z toho osmý antukový. 32letý Mónaco triumfoval v Houstonu podruhé, když v něm vyhrál již v roce 2012. Sock se během zápasu potýkal s bolestivým ramenem a křečemi nohou.[1]
